Aktifitas Sitotoksik Ekstrak Buah Jeruk Pamelo (Citrus Maxima) Terhadap Sel Kanker Servik

Kanker servik adalah jenis kanker yang banyak terjadi di Indonesia. Prevalensi terjadinya terus meningkat, sehingga diperlukan alternative terapi bagi penderita serviks. Potensi jeruk pamelo sebagai agen anti serviks karena pada buah ini mengandung senyawa likopen. Likopen bekerja dengan cara menekan proliferasi atau multiplikasi sel. Pada pasien sel sangat cepat. juga mampu memperlambat bahkan mencegah proses oksidasi dari molekul lain dan mengeliminasi radikal bebas dalam tubuh dapat menyebabkan kerusakan Radikal berikatan terhadap DNA, protein lemak akan merusak fungsi fisiologisnya, gilirannya berkembangnya penyakit servik. merupakan eliminator efektif. Melalui dua mekanisme yaitu maka likopen digunakan Uji aktivitas sitotoksik ekstrak dilakukan metode MTT HeLa. Buah diekstrak maserasi menggunakan pelarut etanol 96%. Kultur (sel HeLa)di ditransfer sebanyak 1x10 4 sel/sumuran media kultur terdiri FBS, penisilin-sterptomisin, amfoterizin-B RPMI 1640 kedalam 96- well plate diinkubasi inkubator CO 2 5% semalam. Selanjutnya pemberian sampel uji seri kadar dibuat replikasi tiga kali (triplo), kemudian diinkubasikan kembali Pengujian hari ke tiga, penambahan reagen MTT, setelah jam terbentuk kristal formazan masih hidup. ditambahkan SDS stoper untuk menghentikan reaksi MTT. Kemudian pembacaan absorbansi elisa reader panjang gelombang 595nm dilanjutkan penentuan nilai IC50. Hasil penelitian menunjukkan IC 50 10.67 µg/µL. hal bahwa memiliki potensi besar. Penelitian memberikan kontribusi pengembangan obat Ekstrak selektif menjadi pilihan penanganan prevalensinya tinggi
